Which is more accurate and why is there such a high difference?

My father was diagnosed with MDS.
Has someone had these mutations or has knowledge about these?
SRSF2(6.12%), TET2(5.19%), RUNX1 (2.5%)
And what so you think about the percentages VAF?
His cytogenetics are normal.
