Hi everyone!

I’m currently running GDF15 ELISAs and had a few questions. No one in my lab has much ELISA experience (at all) so sorry if these are basic.

1. If a sample falls outside the standard curve range, should I determine that based on the raw OD value or based on the calculated concentration after applying the dilution factor?
2. For duplicate samples, what CV cutoff is generally considered acceptable? I’ve seen recommendations of <10–15% for intra-assay CV, but I’m wondering what is typically acceptable (so if I need to re-do a sample, I know which cutoff to use!)
3. How many controls do you usually include per plate?
