South America Itinerary

Hi all, I am going to South America with my Wife and i'm just asking for any feedback about out itinerary and plans and whether we have enough/too much time in all these places. Along with this are there any activities/ sights that you would highly recommend (that are out of the norm/not super well known).
Also, as for the coffee region in Colombia, what would you recommend for transport? Is hiring a car in medellin and dropping it off in bogota a good idea?
Cheers all.
