When you first enter uptown springdale, you can see Katie, Eddie, and Bear in the cutscene walking/running

2 .Give Staminum, Staminum Alpha, or Oddball toadstool to either Rocky Badboya or Mochismo.

after that, the Kupo nut should be an automatic drop. this has worked for me a bunch of times (on both physical copies and emulator)
