What do you think this elongated part in the right side is in intestinal organoids?

Is this black region that I marked looking crypt to you, right? What about the projection? Is villi like structure developing after the addition of my treatment?! I imagine as the lumen of intestinal organoids are small, this projection develops outside the lumen? Or are they converted to cancer?

I have worked with intestinal organoids but I have not seen this sort of elongation like that. What are these in the photo?

What is the black line that appears near the end of the lanes, around the 500 bp region, instead of a white DNA band? I ran the PCR products on a 1% TBE agarose gel. For sample preparation, I mixed 10 µL of PCR product with 2 µL of 6× loading dye and loaded a total of 9 µL per each. What could cause the appearance of this dark band or line? Thanks
