Rate my Missouri road trip itinerary

I live in Chicagoland and am planning to visit Missouri/Kansas Route 66 attractions as well as explore Mark Twain National Forest with my pup over a long weekend. Here is a rough itinerary — any must-see spots I'm missing? Any specific hikes or parks you'd recommend in/neart the national forest? I'm hoping to make this trip in a camper van so I have some more flexibility and am not tied to any hotels at the end of each day.

Night 0

Leave after work, drive to St. Louis area (4 hours)
