‎Today marks a significant milestone in our breeding programme. After years of systematic selection, chemical profiling, and careful cultivation, we are proud to announce that the first seeds from our base breedline are now available for ordering.

‎This is the foundation upon which our future DMT and 5-MeO-DMT cultivars are currently being built—and we're making it available to the community.

‎Our methodology encompasses the systematic selection, controlled breeding, and cultivation of Phalaris aquatica wild germplasm sourced globally. Through Thin-Layer Chromatography (TLC) and bioassays, we identify desirable traits, select specimens with the most promising yield/profiles, and breed them to consolidate and enhance those genetic characteristics.

‎🧬 The Breedline: Three Distinct Chemical Types

‎The seeds now available for ordering represent a base breedline population.

‎This is not yet a stabilized cultivar, but rather the foundational population from which our future cultivars will emerge.

‎Type 1 – DMT-Dominant: High DMT content this is the cleanest chemotype within the breedline population. Here's a bioassay of mine with a clone from this type: https://www.reddit.com/r/Phalaris/s/puYiOuW9fA

‎Type 2 – 5-MeO-DMT-Dominant: High 5-MeO-DMT content, no DMT (often contains a small ratio of 5-meo-nmt)

‎Type 3 – Balanced/Mixed: Contains both DMT and 5-MeO-DMT in varying ratios (no 5-meo-nmt) if you're into mixing DMT with 5-meo-dmt this is the only botanical source that reliably contains both. (a favorite of mine)

‎All three types have been preselected for high yield—there are no low-yielding genetics in this population. If you grow and extract from this seed population, you'll obtain a mix of all three types.

‎If you have access to TLC (or can send single-clone samples for analysis), you can select specific DMT or 5-MeO-DMT clones  with very low to non-existent gramine content. The entire breedline population is already notably low in gramine (strong selection pressure was applied to remove Gramine), with only a minority of outlier clones showing high concentrations.

‎🌍 Help Build a Mediterranean *Phalaris aquatica* Map

‎This spring I've noticed many new members joining, posting photos of grasses in the wild and asking for identification help. It’s great to see the interest! 

‎‎That made me realize : instead of just sharing ID tips, we could actually make it easier for Mediterranean members to find Phalaris aquatica in the field – by sharing verified GPS locations collected during official botanical expeditions.

‎‎I've started gathering this data. The most powerful resource is an interactive map from the Australian Pastures Genebank (APG), which shows the exact collection sites of P. aquatica germplasm from international expeditions (including the 1996 CSIRO-led trips across North Africa and Italy, led by Dr. Culvenor).

‎- The map includes data from the 1996 Mediterranean expeditions (Morocco, Lybia, Algeria, Italy) that are the foundation of many low‑alkaloid breeding programmes.

‎‎1. Open it on a desktop browser (mobile interface can be slow).

‎2. Zoom into your country/region.

‎3. Look for clusters of points – those are areas where P. aquatica has been officially documented.

‎‎🧭 Beyond the GRIN Map – More Data Sources

‎‎The GRIN map is a great start, but we can go further. Here are two other free databases you can search for P. aquatica coordinates:

‎‎- GBIF (Global Biodiversity Information Facility)** – [www.gbif.org](https://www.gbif.org) 

‎  Search for Phalaris Aquatica, filter by “Preserved specimens” and your country. Many herbarium records include public GPS coordinates.

‎‎- Regional Herbaria – Some Mediterranean countries have online herbarium databases (e.g., Italy’s “Herbarium Centrale Italicum”, Spain’s “GBIF-es”).

‎‎⚠️ Ethical & Legal Reminders

‎‎This data is for research and identification only. Do not collect plants without:

‎- Respect for the wild population – in many regions phalaris aquatica is already facing serious genetic erosion due to high grazing pressure and urbanization. don't be part of that problem by over harvesting. Think sustainably. Those wild specimens may have high genetic value for future breeding programmes both entheogenic and agronomic.

‎‎Remember: The goal is to study, preserve, and breed sustainable genotypes – not to exploit wild populations.

‎📚 Quick ID Reminder (for when you get to a site)

‎Use the flowchart from our previous post (I'll work on improving that chart), but here are the three most reliable field traits for P. aquatica:

‎1. Hairs & Auricles – None. If you see fuzz or little claws at the leaf base, it’s not Phalaris.

‎2. Green Stripes on Panicle – When the head is still green, all Phalaris species have alternating light/dark green stripes. No stripes = not Phalaris.

‎3. Sterile Lemma – P. aquatica has one single, thin, long, pointy sterile lemma at the base of the seed. Short, thick, white lemmas = P. truncata* or P. brachystachys.

It's not much but there's a story twist

25mg dry crushed leaf sample was added to 1.5ml centrifuge vial. 0.3ml 25% ammonia and 1ml chloroform was added and vial shaken periodically for 5 hours. Chloroform fraction transferred to another vial and left to evap slowly and that is the result.

That's DMT freebase crystals in case you're wondering. Leaf sample is from a phalaris aquatica hybrid clone from our selected DMT breedline: https://forum.dmt-nexus.me/threads/the-nexian-phalaris-breeding-programme.369757/post-3937813

The same clone was analysed by thin layer chromatography (TLC) and shows a clean DMT profile (hardly any gramine detected just a small amount of NMT).

First DMT clone bioassay of the F2 hybrids

Following the transplanting of 750 F2 hybrids and 10 North African accessions, a preliminary bioassay was conducted on two high-yield DMT clones. These clones were selected from the hybrid plot based on Thin Layer Chromatography (TLC) analysis, specifically targeting non-methoxylated alkaloids.

‎- Material & Extraction: The plant material consisted of 8.5g of fresh leaves, which were subsequently dried to a final weight of 2.000g as determined by a 0.001g moisture analyzer. The extraction method involved boiling the dried leaf material twice in distilled water with citric acid, each for 30 minutes. The resulting solution was then basified with NaOH and extracted twice with chloroform. The chloroform pulls were combined and evaporated in a pre-weighed 50mL beaker. The crude extract yield was 18mg from 2g of dry leaf, representing a 0.9% crude yield (w/w dry weight). Based on dose-response, the crude extract was estimated to be approximately 50% DMT, resulting in a net DMT yield of approximately 0.3–0.4% of dry leaf.

‎Pic 5, 6 is the TLC analysis result of one of the two clones tested. Clone sample is lane 7. Lane 4 is MHRB. All lanes on the plate are 25mg dry weight samples soaked in 1ml chloroform + few drops of 25% ammonia. Notice the two largest blue fluorescing spots at the top (DMT RF) are the MHRB and the DMT clone (which based on it's DMT RF fluorecence intensity seems to be roughly 40% that of MHRB DMT spot, which is backed by the practical crude yeild extracted.

‎Pic 3 and 4 are the selected clones being transplanted for clonal propagation.
‎Pic 5 is the 8.5g fresh leaves extracted which gave the 18mg crude freebase.

‎Bioassay Results: The bioassay revealed that the onset, peak, duration, and subjective effects were strikingly similar to that of DMT extracted from MHRB.

To be absolutely sure i pulled out my Stash of MHRB extracted DMT freebase and vaporized some.. it only lead to increased intensity of the same effects i had from the hybrid clones extract. I wish i smoked the clone extract at higher dose all at once... The room was just starting to morph when i reached the peak of the last dose and had no more left to smoke.

I was smoking the. Rude freebase in incremental doses to explore it. Now that i am familiar with these selected clones benign qualities my next harvest i will go for breakthrough.
