First timer, thoughts?

I'm curious to get some opinions and knowledge dropped on me. This is my first attempt and first time using agar other than microbiology class. Spores came from an spore print (2022) of a wild species. Plates inoculated 15 days ago. I'm planning on doing the first transfer ASAP to get rid of contamination. First I wanted see what the group mind thought about what's going on with these plates.

Want help getting rid of black mold

Its been frustrating, but my last 9 jars have been continously infected either by trichey or by black mold (mostly black mold). I tried different grains (millet and brown rice) and I even tried different methods such as adding chalk powder to the jars. I need help figuring out what mistake am I making?! I usually put half plate of agar into 250 gms of grains(I have 550ml jars), and i think that's the culprit as it all starts to deteriorate either from the agar or the grains near the agar which have turned soggy or have swollen. For the first 3-4 days I usually see the mycelium taking over the top layer making a thin fluffy layer, and suddenly on the 4th of 5th day I notice greyish spots which spread quickly in 24 hours :(( What can I do next time, not put the agar pieces but just scrap the myc on top and inoculate that?

TLDR: need help with black mold or whatever contam im getting.

Help saving my mycelium :d

A month ago, I inoculated these Petri dishes with Golden Teacher spores from a vial, intending to create a liquid culture.

However, over the last two weeks, the mycelium seems to have stopped growing or is growing very slowly whereas it should have been almost fully colonized by the three-week mark. Additionally, some areas of the mycelium have started turning brown.

I cut out a piece a week ago to start a new culture; perhaps it got slightly contaminated then, or maybe some alcohol dripped onto it, though I doubt that.

I just wanted to ask if there is actually something wrong with my plates, if there is anything I can do to strengthen them, and if I should still proceed with my plans to use them for liquid culture.

Question for newbie working with agar

Hello,

I have these two agar plates i transfered and kinda forgot about lol.

Whats best to do with these? Make transfers and what to do with the rest of the plate? Put it to grain and pray it works out or what do you guys do with the rest of the plate after a transfer?

If transfer, where would you take from?

Really new to working with agar so just looking to learn as much as possible.

Grain To Liquid Transfer

I’ve tried googling, and skimming through here but I haven’t found anything. Has anyone had luck using colonized grain to make a LC? I have some Golden Teacher i want to make a LC of but don’t want to get Petri dishes, and agar. What’s the success rate for using grain, and do I just grab a clump and drop it in same as I would normal mycelium?

No pour agar - Can I fully close these lids or do I need to open it slightly or modify the lid during sterilisation?

First time doing agar plates and wanting to do the no-pour method. I received these pp5 condiment cups with the lid attached to the cup, and the description says "leak-resistant storage" when the lid is closed.

I'm planning on using my Instant Pot Duo for sterilisation where it's max 12 psi. If I use the "pressure cook" setting on high for 45 min and leaving it in there to cool for 2-3 hours, will that be enough?

Would I be fine fully closing the lid when I put it into my instant pot for sterilisation? Would I need to wrap it with parafilm, drill a hole into it and cover with micropore tape or keep it opened slightly? I found when I keep it open slightly, there's a bit of opened gap so I'm not sure if it would be fine

Is this agar appropriate

I finally found an agar with cheap shipping for cultivating my seeds then i thought this isn't specificily for orchids and im new to growing from seeds so here is a list of the ingredients. soy-peptone, micro, and macro nutrients, will this work for flasking or does anyone know where i can get cheap agar.

Clean for grain ?

I'm newbie. Cloned from tissue. Can't beat the condensation. Few transfers all had bacteria. Then put in non nutritional agar only and came out rhizomorohic . The last transfer agar and malt , dish been upside down since agar hardened. Paranoid about bacteria. Seams clean ?

Wanna transfer to grain .

Fruiting from plate

I’ve seen this happen to other people before, but this is the first time I’ve experienced it. Do I let it grow? Should I try to take a flesh sample and clone it?
What exactly does it mean when the mycelium wants to fruit on a plate? Is it a sign of healthy genetics?
I had previously cut into this plate to try to isolate some good looking growth and decided to keep it going in case I needed to cut more from it.

Is this worth adding to grain?

I recently caught the agar bug and have been doing some experimenting. I purchased an agar culture and did some transfers from it. I left it sitting in my SAB for 2 days and came back to the purchased agar plate fruiting. I decided to remove 4 of the fruits and place them on an agar dish. This is how they are looking after about a week. Would this plate be worth transferring to a grain bag? I believe the only discoloration I am seeing is the fruits themselves, but I could be very wrong! Any information is appreciated!
