Red hue might be the reflection of a face I am not sure but Spider-man The Lost Hunt had a similar image for Peter's eyes.

It was when he was intoxicated and was devoured by his inner conciousness giving into "the Spider". After years of trauma his mind was pushed to think of himself as two parts of a whole: the man and the spider. He made himself believe the "the spider" was the cause of his suffering... at this time he was traumatized, struggling and he was pushing his loved ones away... when he was going through it alone, "the spider" managed to devour him with the help of the toxin. It was Mary Jane who succeeded in reaching out to him by telling him, he is not alone.

I am not saying they will adapt pieces from this comic story, but having black&red eyes as Peter becomes more of a spider during a time of sadness and isolation might have been a insipration for the movie aesthetic.
