Membrane overnight incubation with primary antibody with NO SHAKING/ROLLING at 4 degrees?

Hi all, we do not have a cold room in our lab. Do you think i could incubate my membrane fully covered in TBST/primary overnight in the 4^(o)C fridge without agitation? Has anyone done this before and how did it turn out?

10x TBS buffer floating precipitates? What is this?

This 10X TBS buffer i prepared from scratch is around 1 month old. I just noticed that they have this floating things. The solution is otherwise clear. Anybody has any idea what this is or if it is still useable? Thank you!

Whats your least favorite/most hated thing to do in the lab?

Mine is preparing buffer... converting how much to weigh and adjusting the pH is just.. so painful. I dont mind doing all sorts of extraction and running 10 qPCRs. But if u ask me to prepare any buffer at al....

HEK293 cell lysis for protein extraction rant. Is it okay to still have blob/small clumps in samples before centrifugation?

Just did my first protein extraction and oh my GOODNESS.. i crashed out 5 times.

For context, i am extracting HEK293 72 hours after transfection (so probably 1mil cells total?). The protocol from my senior said to use 100uL/well for 12-well plate, so i did that. and GOSH it was so difficult to resuspend. There is this blob/goo (likely DNA based on what ive read) that keeps getting clogged in my pipette tip and it makes pipetting create so many bubbles/froth UGH! Even when i use 1mL syringe and 21G/25G needle to shear it, it clogs the needle as well,,,, After like 10 times of moving the syringe up and down, i can still see this blob albeit broken up. Just looks like mucus all over the sample.. I gave up and just spin it down. The pellet i get afterwards look transparent, not sure if this is correct? usually i get white cell pellets when i spin the HEK293 down for FACS.

Do you think i can just not shear this blob/goo and fish it out? i.e. when it gets stuck in the needle i just remove it altogether.

I would really appreciate any tips and tricks on WB as well, thank you everyone.

P/S i froze my samples in -80, not looking forward to the rest of WB. Already encountered issue on the first step (supposedly easy) of WB...

The Red Oolong Tea being my favorite so far! Heard so many good things about this bar and it did not disappoint, wish it could be smoother though.

The autumn bar in the middle was unfortunately a let down, it was very grainy :(

Do you guys have any other suggestions for Taiwan chocolate? Thanks all !

Got this from Cheers at 5.40 (for 275g pack)

Tasted pretty okay, a solid 6.5/10 overall.

● The nasi lemak is not very coconutty, has ginger bits, so it tastes somewhere in between nasi lemak and chicken rice.

● The portion is so small... there are like 4 cubes of chicken :( but rendang taste is decent and pretty flavourful.

● The sambal is more on the sweeter side, which i like

How do you guys like this?

Idk who might need this..but i just wanted to let you guys know that the seal is still usable with brown/yellow old seals (i used microseal 'B' from BioRad). I freaked out after i pasted it on and it was really brown (browner than in the photo here) but since i already added everything i just went ahead with the run. The results were fine, technical repliates are consistent and ct values are similar to my previous runs. Cheers
