How long should I wait?

Hi, I took a spore print from a "Mexican" Freshroom grow kit. I then used a sterile cotton pad to transfer it onto an agar plate. From the cleanest plate, I transferred sections onto a second agar plate. The image you see now show the results from those second plates. Can I take pieces from them now and transfer them into LC jars? Or do I need to go a step further and transfer them onto a third agar plate? Sorry—I'm still pretty new to this! :)
