Do skills like, matter, for a casual playthrough of this game or do i need to seriously think about this?

Just to be clear, Im just wondering if the skills are more like an addition to the gameplay or if its more of a "Hey you need these to do well" kinda deal. And if so, which are the good ones?

Is this a reference to BL1? Is it a thing he says often? Or is this a one-off joke?

Im specifically referring to the part at the end where he mentions "Wall-Sphincters". Like, is that a joke im not getting or is it something that comes up in Bl3? Or is it really just a one off.
