LogD shake-flask procedure.

I have been following the method presented in this paper and according to it, since I have a very lipophilic compound I should follow procedure 1b or 3. I am trying to replicate 1b but I have a few questions regarding the meaning of the dilutions and the Vw / Vo ratio. For example, If I want to do a trial with a Vw / Vo ratio, which procedure would be correct, 1ml of water and 9ml of octanol, or 1ml of water and 10ml of octanol?. If I do the second one, my total volume would be 11ml and if I also want to do a Vw / Vo ratio of 1 and a Vw / Vo ratio of 10, should I aim for a total volume of 11 as well? or could I do 10 ml for the ratio of 1 (5 and 5 ml each) and then again 11ml for the ratio of 10 (10 and 1 ml respectively?.

Additionally, I have a stock solution in DMSO that I can dilute in PBS as the water phase, when the procedure asks for a 100x dilution, is that considering only the water phase volume as the final volume? or is it considerinf the total volume as the water phase + octanol volumes together? Since the water to octanol ratio changes, if I dilute my sample in PBS (water phase) for the Vw / Vo ratio of 1, the amount of sample would be different than the ratio of 0.1 with less water phase and different than the ratio of 10 with more water phase present. On the other hand, If I take the 100x dilution as related to the total volume of water + octanol, also the amount of compound would be different depending on the trial because some will have a total volume of 11ml and other of 10ml.

In summary, what should I keep consistent? total water+octanol volume? sample concentration? niether? Help please.

¿Como pagar gastos comunes en el extranjero con el mejor tipo de cambio?

Me voy a ir por mas de dos meses a Canadá y no se cuál es la mejor forma de pagar mis gastos diaros. Cuando me he ido de viaje antes llevaba dolares en efectivo y con eso pagaba mis gastos de comida, transporte, etc. Pero, eso servía cuando me iba máximo una semana. Ahora que me iré un tiempo considerable, quiero saber cómo conviene pagar estas cosas más comunes. Cosas como insumos del supermercado, transporte en bus, lavanderia, etc.

How many surfers does it take to stop a wave? [Request]

I guess that in order for a surfer to move forward on a wave, they should be taking energy out of the wave itself. If that is so, then how many surfers does it take to take away the energy of a whole wave? For maths sake imagine that the wave us in the middle of the ocean, it could be circular or a straight line, as you wish, and the distance the surfers travel is 100m or 1km, whatever is most convenient. I don't know in the slightest what type of math should be applied here, but I was just curious.

Housing options for a limited stay

I am going to USAK for an internship for a total of two months during the summer (mid July, all of August and the start of September). I was looking at housing options in Airbnb and they range from 1000 to 2000 CAD. However, I wanted to ask you as locals if you think there are cheaper options.

How do you fly on a plane with your research compound?

I am going to fly to Canada to continue my research during an internship there. The thing is that I have to take with me my synthesized compound in order to do more analysis with their tools. My thesis advisor told me that it is normal to go on a plane with your compound in your luggage as long as you have the proper documentation for it.

Has any of you donde something similar? Who emits the documentation and what does it say? I am extra worried because I will be going from Peru with a stop in Mexico, and my compound will be a beige powder on a vial, so... super suspicious. how do I avoid a cavity search? 😞
