Agar contam?

Hi,

Fairly new to growing.

GT LC grown on agar. Most of the plates are largely tomentose with normal looking patches of rhizo.

Seeing spiky type growth in a couple.. Not sure if it's contam or a weird rhizo?

Photos attached.

Trich trouble

Hey, I've been trying to grow for about 3-4 months and I've had so many trich problems.

Literally every S2B seems to go ok, then about 2-3 weeks in trich pops up before fruiting happens.

My process is evolving. Started broke boi tek, then pressure cooker. Then made LC and tested it on agar, currently growing jars from proven agar.

But even the past jars from the LC that's not contaminated haven't been workIng for S2B.

I've done a lot of research, and I'm careful. I can't understand it.

Current method:

Using brown rice in 1l/1qt jars. Filter disc, and self healing injection port on jars.

Innoculating jars in a pop up SAB. Set it up in advance. Spray the crap out of it with 70% isopropyl. Leave it for a few hours. Spray again before innoculating.

Spray the jars before innoculating.

Was innoculating with LC. The LC has now spawned 20-30 agar plates no contam.

Now moving to agar but haven't bulked any yet.

Using fresh sterile needles and syringes.

Jars are all ok. If contam I don't S2B them. All fully colonized, no uncolonized grain.

When going S2B I use a tub, not modified, loose lid no seal.

Use field capacity 100% coir, 1:1 or 2:1 ratio. No added ingredients to coir.

Coir originally pasteurized using bucket tek.

I know there are different opinions, but because I've had so many problems, I now err on the side of caution.

Use gloves, mask, and spray with isopropyl. Pre spray tubs, and also give a wipe down before spawn. Spray gloves before using.

I don't touch the jars anymore or use a spoon, just break and shake into tub.

If spawning more than one tub, I change gloves in between.

I have just started pressure cooking the coir as a precaution because I don't know what else to do.

When spawning, I've tried (pseudo) casing, no casing.

I've tried spraying and no spraying before closing lid. If spraying, I use water that has been boiled twice and then cooled.

I've cracked the lid to go straight to fruiting, and tried neglect tek.

Done about 10 S2Bs and none have worked. They all get trich at some point. The first few, I get it. The next few... maybe, but all of them?

I have no idea what to do or why it keeps happening?

The next batches will be from agar to grain with pressure cooked coir, so I'm hoping at some point something will work.

Am I missing anything, or is there anything else I can do?

LC question

Hi, newish to this thing.

I've made up some LC but forgot to shake them for a while and there are large clumps of mycelium on the surface.

Is this an issue at all? Is there anything I should do?

FAE question

Hey, I'm new to all this. Haven't had a successful grow yet.

I've spawned some grain to bulk in some cheap plastic boxes.

They are 20 litre plastic boxes that are about 26cm high. I picked them thinking that it would provide a more stable microclimate.

All is going well, condensation looks good etc.

My question is really about FAE.

The lids are loose lids like shoe box lids with no seal, which should provide enough passive FAE, but there is about 15-20cm from the top of the substrate to the top of the box.

Is that ok do you think?

I'm thinking about c02 pooling/concerned there may not be enough FAE because of the height.

Should I add air holes closer to the substrate?

This is frustrating but I will solve this puzzle

I started with broke boi tek with a mix of spore syringes and LC syringes.

I read everything carefully and watched a bunch of videos and learned a lot.

First 4 jars I spore shot got contaminated. Binned.

Ok, I kept going. I made a bunch more jars using LC and also made some LC jars. The LC jars seem ok.

All of the grain jars I have taken to S2B have been contaminated with trich at some point after bulking.

I try the cut out and quarantine method and put it outside with each box. It comes back. Optimism is great but experience is better. I learn the hard way about trich myself when I could have just listened to what everyone says.

I buy a pressure cooker, buy more gear, buy glass petrie dishes.

I do a test run of agar, muck it up, too much condensation, growing nothing much. A couple grow mould.

I try again, very strict cleanliness/sterility, using home made SAB. The agar plates seem to be going ok. No contam, and it confirms my homemade LC is ok.

Pressure cooked jars inoculated with LC all look good.

About to spawn my first pressure cooked grain. Crossing my fingers like mad. I still have 2 S2B boxes from before the pressure cooker that might survive but we'll see.

I will crack this nut one day. The puzzle solving is weirdly almost as good as trying to get the results.

Learning a lot along the way.

Contamination or bruising?

This is my first spawn to bulk.

Do you think it's bruising or contamination?

It smells fine - nothing odd.

In real life the colour is hard to gauge. Could be blue?

It's winter here so there has been some condensation build up over that area particularly at night, with some heavy droplets building up in a patch on the roof above.

Possibly why it's first to colonize, but is it possible the water droplets are causing bruising?

Or am I just delusional...?
