Santa Cruz, Miraflores or Asuncion?

I'm seriously thinking about leaving the UK and making a move to South America.
At the moment, I'm looking at Santa Cruz, Asunción and Miraflores. Because of visa requirements, I'd likely split my time between Spain and South America every few months.

For anyone who's lived in or travelled to these places, which would you recommend, and why? Are there any other cities I should be considering?
I'd really appreciate hearing about your experiences before I make the move.
