The first three pics are different angles of what im assuming to be a jade leaf callus? I just cut it up today to propagate onto new media and upon cutting it reminded me of broccoli.

Last two pics are of a Lophophora species, a type of globular cactus, and it grew some roots and an off shoot of some kind along with some weird underdeveloped stuff near the center.

These were put on the same media using a 1:1 ratio of NAA and BAP. Just curious as to what kind of growth this hormone ratio produces, I know it varies among species but is there any general trend?

Any input/advice is appreciated!

Soxhlet Update!

I recently posted a timelapse of a soxhlet extraction of SS02 x eileen, and I'm back with more notes and comments on the process and as to whether its helpful or hurtful.

I ran some Juuls, and unless there was some super wide variation in the cuts (one was quite larger/older than the rest) I can say that the soxhlet was about half as efficient as the french press method. I will continue to do more refined tests as well as tightening the SOP for running a Cielo in the soxhlet.

yield for 69 grams of cactus was 1.14 via french press

yield for 66 grams via soxhlet was 0.5

I think the lack of agitation with the plant matter leaves the mescaline stuck in the plant matter leading to a less efficient extraction. I used 500 mL of solvent in the soxhlet and 1000 mL via the french press. French press from start to finish takes me about 2.5 hrs and the soxhlet takes 1 Hr for the first cycle and 40 min for the ones following. So I can get roughly 4 cycles on the soxhlet in the amount of time it takes to do the french press.

If this method is refined to determine how many pulls are needed to extract all mesc then this could be an easy way to have the extraction running in the background while tending to other projects not to mention not breathing in solvent, and reduced solvent usage saving money and health.

I'm open to any ideas or suggestions on ways to improve the extraction TEK for the soxhlet!

Sso2 x Eileen

So Ive been running smaller "test" batches for cultivars in my soxhlet as 50-80 grams of dry material fits nicely.
Just curious if anyone has ran this cultivar and what their yield for citrate was like, mines at abt 2%!

Edit: For all wondering, this is a Soxhlet extractor, it boils solvent in the lower flask and the part thats out of view is a coiled condenser that redistills fresh solvent over top of the plant material. This allows me to do as many "pulls" on the plant matter as I want and I can set this up and walk away which frees my time up as I'm usually pretty busy.

Question around oxidation

I have found evidence that cytochrome P 450 converts n,n-DMT to DMT-NO but I can't find any literature supporting that this happens from the air as well. Im assuming when DMT is left in the air long enough the oxidation product could be DMT-NO and I think I could covert it back to regular NN via dissolving in an acidic solution such as vinegar followed by adding zinc powder to reduce it. Filter said solution to remove excess Zinc powder and then free base it by adding base. I'm open to any advice as to why it would or wouldn't work.

Quick question!

If I were to run a soxhlet extractor with ethyl acetate and use magnesium sulfate to keep any water out of the solvent, could there be any adverse reactions from heating/boiling the solvent with the magnesium sulfate in it?

Trying something different

So I recently got my hands on a soxhlet extractor and tried to use it for cielo. I think I may have over hydrated my cactus paste or maybe water was leaking in because I didnt grease the joints. Anyways now I have to get rid of the water to be able to finish up and im not too sure how to go abt it. Open to suggestions and questions !
