I've got multiple maxed out characters, but some rewards are locked behind "mastery?" What's the difference, and how do I level up each one?

Just wondering which characters should absolutely never be in the front.

Personally, my main teams are Mark, Beast, Titan, and Anissa, Lucan, Conquest, each in that order. I'm assuming Mark and Anissa as leads are fine, but would any of the other be better?
