Disclosure: I do content work for a longevity supplement brand, so please weigh that accordingly. I’m interested in the research side of longevity and try to look at the evidence before jumping to conclusions.

There’s a statement you see constantly in longevity discussions:

“NAD⁺ declines with age.”

But how strong is the evidence behind that statement?

I came across a critical review that specifically examined this question:

The authors looked at studies examining NAD⁺ levels across different species and tissues, including humans.

What caught my attention was the difference between how confidently the idea is often presented online and how limited the underlying evidence can be.

The review points out that:

• Studies have examined different tissues and species rather than one consistent biological system.

• Results aren't always consistent between tissues.

• Human data are particularly limited.

• Some studies involve relatively small numbers of participants.

• Much more research is needed to understand how NAD⁺ levels change across different tissues throughout aging.

So the interesting question isn't necessarily:

“Does NAD⁺ decline with age yes or no?”

It may be:

“Where does NAD⁺ change with age, by how much, in whom, and under what conditions?”

That distinction seems important.

It also makes me wonder how much of the current NAD⁺ conversation is based on evidence from specific tissues or animal models versus what we actually know about normal human aging.

What happens to the immune system when researchers target mitochondrial quality?

Disclosure: I do content work for a longevity supplement company, so take that into account when reading this. I’m sharing this because I found the underlying human study interesting, not as a product recommendation.

I came across a recent randomized, placebo-controlled trial looking at urolithin A and immune aging, and I thought the study design was worth discussing.

The paper is:

“Effect of the mitophagy inducer urolithin A on age-related immune decline: a randomized, placebo-controlled trial”

What caught my attention is that the researchers weren't simply measuring a general wellness outcome.

They looked at immune-cell populations and immune metabolism, including different CD8+ T-cell subsets, mitochondrial-related measures, and other immune markers.

The study involved 50 healthy middle-aged adults, with participants receiving either urolithin A or placebo for 4 weeks.

That makes the paper interesting to me because it raises a broader question about how we think about "immune aging."

We often talk about aging as if it's one process happening uniformly throughout the body.

But the more aging research I read, the more interesting it becomes to look at specific cell populations and cellular processes rather than relying on one broad aging label.

And this is where the study gets particularly interesting.

The researchers reported changes in several immune-cell characteristics after the intervention, including changes involving CD8+ T cells and mitochondrial-related measures.

I'm curious how people here interpret this kind of research.

I was simply reading this review and thought it raised an interesting question about why NAD⁺ appears across so many areas of aging research.

NAD⁺ seems to appear in almost every major discussion about aging biology.

The review describes its involvement in areas such as:

Question: What's the most promising longevity breakthrough you've seen recently?

I've been following longevity research for a while, and it feels like the field is moving faster than ever.

One breakthrough that really caught my attention is partial cellular reprogramming a technique that aims to make older cells behave more like younger cells without turning them into stem cells. After years of animal studies, the first human trials have now begun, making it one of the most closely watched areas in longevity science. Researchers hope it could eventually help restore tissue function and slow aspects of biological aging.

Research on Creatine and Brain Energy
